The endocrine effect almost nobody mentions
Melatonin is a hormone, and hormones do hormone things. A 2025 randomised study in 16 intact male Pomeranians with alopecia X gave melatonin at 0.1, 0.3, or 0.5 mg/kg twice daily for 45 days and found dose-dependent changes in sperm concentration and motility, along with a reduced testosterone-to-estradiol ratio (Veterinary Medicine and Science, 2025, doi:10.1002/vms3.70410).
For a neutered pet dog this is a footnote. For an intact male in a breeding program on long-term melatonin, it is a genuine consideration worth raising with your veterinarian — and it is essentially absent from consumer articles on this topic.
Xylitol: the part that turns a supplement into an emergency
The Merck Veterinary Manual notes that the dog is the only domestic species in which xylitol toxicosis has been reported. The thresholds it gives are specific: above 100 mg/kg (99.2 mg/kg), xylitol triggers dose-dependent insulin release and profound hypoglycemia; above 500 mg/kg (500.4 mg/kg), the risk shifts to severe liver failure (Xylitol Toxicosis in Dogs, revised September 2024).
Run those numbers for a small dog and the margin disappears fast. A 4.5 kg (4.5 kg) dog reaches the 100 mg/kg threshold at roughly 450 mg of xylitol, and the liver-injury threshold at roughly 2.25 g. That is straightforward arithmetic from Merck's figures, not a measurement of any particular product — but it shows how little material is involved.
Timing matters as much as amount. Hypoglycemia can appear within 30 minutes or be delayed 12 to 18 hours depending on what the xylitol was formulated with. Liver enzymes often start climbing at 4 to 12 hours, while visible signs of liver injury may not surface until 24 to 48 hours. A dog that seems fine at bedtime is not necessarily out of the woods. - Scan for xylitol under all of its names. It appears as birch sugar, birch sap, birch bark extract, wood sugar, wood extract, E967, xylit, xylite, xilitol, or meso-xylitol. Any one of these means do not give it to your dog.
- Check whether melatonin content is stated per unit. Per tablet, per chew, per millilitre. "Proprietary blend" means the number is hidden.

The second problem: the label may not be accurate
Even a xylitol-free human product carries a dosing problem. Testing of 25 melatonin gummy products sold in the United States found 22 of 25 (88%) inaccurately labelled, with measured melatonin ranging from 74% to 347% of the declared amount; one product contained no detectable melatonin but 31.3 mg of CBD (Cohen PA et al., JAMA 2023, PMID 37097362).
Earlier work found the same pattern across formats. Analysis of 31 supplements from 16 brands found melatonin content from 83% below to 478% above label claim, lot-to-lot variability within a single product of up to 465%, and serotonin detected in 26% of products tested (Erland LAE & Saxena PK, Journal of Clinical Sleep Medicine 2017, doi:10.5664/jcsm.6462).
