Amantadine for Dogs in the UK: Off-Label Dosing, Vets and Timeline

In the UK, amantadine is prescribed for dogs off-label from a human prescription medicine — no amantadine product is authorised for dogs anywhere in the world. Dosing is typically 3–5 mg/kg by mouth, it is added to an NSAID rather than replacing one, and the only randomised placebo-controlled trial in dogs found the benefit at day 42, not at day 7 or day 21.

If your vet has just raised amantadine, your dog is almost certainly at the stage where an NSAID alone is no longer holding the pain. Here is what the evidence supports, what it does not, and the UK specifics that change how the prescription reaches you.

How off-label prescribing works here

The UK Veterinary Medicines Directorate authorises veterinary medicines; it has never authorised an amantadine product for dogs, because none exists. A vet registered with the RCVS may nonetheless prescribe a human medicine for an animal under the prescribing cascade, and that authority comes from the Veterinary Medicines Regulations rather than from a product licence. Amantadine is a human prescription medicine, so it is dispensed against your vet's prescription and never sold over the counter.

Two UK consequences follow. First, a high-street pharmacy may not stock it, and you may be supplied a compounded liquid instead of capsules — and compounded concentrations differ between pharmacies. Check the mg per mL on every new bottle. A syringe mark memorised from the last batch is the classic route to a serious overdose in a small dog. Second, if you are rural, ask whether a 6-week supply can be dispensed at once, because this drug should not be judged — or interrupted — at two weeks.

What amantadine does, and why it needs a partner drug

Amantadine is a weak NMDA-receptor antagonist. After months of joint pain, spinal cord neurons become progressively easier to fire — central sensitisation, or wind-up — and NMDA receptors drive that amplification. Amantadine dampens it. An NSAID such as meloxicam for dogs in the UK works at the joint on inflammation; amantadine works on the signal in the spinal cord. Different target, different job. That is why a correctly dosed dog can still be sore, and why the two are combined. Gabapentin for dogs in the UK belongs to the same central-acting group.

The 2022 AAHA Pain Management Guidelines (an American standard) treat chronic canine pain this way on purpose — stack mechanisms rather than push one drug higher (Gruen et al., JAAHA 2022;58(2):55–76, PMID 35195712).

The evidence, and the six-week rule

Lascelles and colleagues enrolled dogs with hind-limb osteoarthritis pain already refractory to an NSAID, kept them on meloxicam, and randomised them to amantadine 3–5 mg/kg once daily or placebo. Owner-scored activity improved significantly on day 42, with no significant difference at day 7 or day 21 (J Vet Intern Med 2008;22(1):53–59, PMID 18289289).

Canine pharmacokinetic work found rapid elimination and suggested every-12-hour dosing may suit dogs better than once daily (Norkus et al., J Vet Pharmacol Ther 2015;38(3):305–308, PMID 25427541). A 2025 study reported it as an option for neuropathic pain in dogs with degenerative lumbosacral stenosis (BMC Vet Res 2025;21(1):469, PMID 40671053).

Not supported by evidence: amantadine as a stand-alone painkiller in dogs, any effect on cartilage, and any slowing of arthritis progression.

Amantadine beside Librela

UK dog owners meet a naming difference here that trips up anyone reading Australian articles. Side effects

  • Common, mild, usually early: soft stools, diarrhoea, wind, restlessness or agitation, sleepiness that fades.
  • Less common: increased thirst, difficulty urinating, faster heart rate.
  • Call your vet: seizures, tremors, incoordination, marked agitation, limb swelling, repeated vomiting.

Raise kidney disease before the first dose — amantadine is cleared renally. Raise any serotonergic medication too, including tramadol for dogs in the UK and behavioural drugs.

What you control long before amantadine

Every dog in the amantadine evidence base had already failed something.
